Patent 5021550: Method for preventing deletion sequences in solid phase synthesis
Solid phase synthesis has already been used to prepare the P2 fragment which has been purified to as high as 90% of the activity present in the natural P2 fragment.
Solid phase synthesis has been known for some time for use in the preparation of peptides and other oligomers such as nucleotides.
A major disadvantage of solid phase synthetic methods for the preparation of oligomeric materials such as peptides and nucleotides results from the fact that the reactions involved in the scheme are imperfect; no reaction proceeds to 100% completion.

Solid-Phase Synthesis of 1,2-Benzophenazine and Some Fused Imidazole Derivatives
Thus, the use of the solid-phase method for the synthesis of heterocyclic compounds may significantly simplify preparation of such compounds and provide for the synthesis of derivatives, which cannot be obtained in solution.
Thus, we have found that the synthesis of benzo[b]phenazine (1) occurs in 98% yield upon treating a mixture of 1,2-phenylenediamine (lla) and 2,3-naphlho[b]quinone (III) in a vibrational ball mill.
The synthesis was carried out analogously to procedure A but using silica gel particles as the working element, mainained for 24 h, treated in VP mixer with steel balls for 2 h.

 Solid Phase Synthesis
Solid phase synthesis has found praise in its ability to separate the resin-attached targets from large amounts of excess reagents and impurities.
Linker groups are attached to resin beads in the solid phase and the reactants are passed over in solution to make the intermediates and a final detachment-from-resin reaction is run to get the desired product out of the column.
But this area is still growing with respect to isolating target molecules that are smaller than the peptides the synthesis was originally developed for.

 Solid phase peptide synthesis
In solid phase synthesis, the C-terminal residue is attached covalently to an insoluble resin.
A very high efficiency is needed for amino acid synthesis to work properly, since any mistakes made are cumulative.
As each new residue is added, the elongation product attached to the solid resin is recovered by filtration (or centrifugation).

 Solid phase synthesis
The revolutionary principle behind solid phase peptide synthesis (SPPS) as conceived by Merrifield in 1959 is that if the peptide is bound to an insoluble support then any unreacted reagents left at the end of any synthetic step can be removed by a simple wash procedure, greatly decreasing the time required for synthesis.
The field of solid phase peptide synthesis has rapidly grown since its inception by Merrifield but is dominated by use of BOC/benzyl and Fmoc/
The traditional resin used for Merrifield synthesis was a chloromethylphenyl substituted resin.

 Barany Abstracts
Incorporation of unprotected phosphotyrosine during the synthesis of peptides with multiple phosphotyrosine residues has been studied as a function of coupling conditions and the absence or presence of intervening amino acid residues.
The pyrophosphate-forming side reaction is more severe with increased coupling dimes and/or repetitions of coupling and occurs only when the phosphotyrosine residues are directly adjacent to one another.
However when it is used to incorporate consecutive phosphotyrosine residues, a pyrophosphate linkage can form between the two adjacent tyrosines.

John Wiley and Sons, Inc. is pleased to announce plans to publish Solid-Phase Organic Syntheses (SPOS), a new book series that will provide organic chemists with verified solid-phase synthesis procedures that can be repeated in the laboratory.
Solid-phase quench methodologies for library generation, while of obvious value, are not considered appropriate for this series; however, the preparation of the resins themselves may be.
Though focusing on library synthesis procedures, other issues addressed may include resin preparation, library screening and encoding.

 ScienceWeek
Toward that end, the authors have synthesized an environment-sensitive fluorescent amino acid, Aladan, and incorporated it site-specifically into proteins by both nonsense suppression and solid-phase synthesis, and the authors have used it to probe the electrostatic character of the B1 domain of streptococcal protein G (GB1) at multiple sites by steady-state and time-resolved fluorescence.
These materials are formed by dissolving a salt (for example LiI) in a solid host polymer such as poly(ethylene oxide) (2-5), and may be prepared as both crystalline and amorphous phases.
The electrical, optical, and electrochemical properties of conjugated polymers can be modified by chemical synthesis and are strongly affected by relatively small perturbations, including changes in temperature, solvent, or chemical environment.

Development of synthetic tools for the solid-phase synthesis of CNS active compounds
The researchgroup at the university of Amsterdam will focus at developing novel N-acyliminium ion chemistry on solid phase, so that new heterocyclic compound classes (with possible CNS activity) will become available via solid support reactions.
Despite greatly expanded research efforts in CC in the past three years, the development of new techniques for the synthesis of large numbers of compounds will also be vital in the near future.
Combinatorial chemistry (CC) has recently become a major topic in organic synthesis as a result of its potential to develop into an essential tool for the discovery of new pharmaceuticals or catalysts.
