As the name implies, Southwestern blotting is a variation of the Western blotting technique.
These data suggest that the positive broad spot detected by Southwestern blotting is a cluster of several different proteins.

Southwestern blotting, based along the lines of Southern blotting (which was created by Edwin Southern), involves characterizing proteins that bind to DNA.
The proteins undergo gel electrophoresis and are subsequently transferred to nitrocellulose membranes similar to other types of blotting.
For other types of blotting, see Northern blotting, Southern blotting, Western blotting, and future blots for analysis.

Southwestern blotting, first described by Bowen and colleagues (1), is a powerful technique for identifying and characterizing DNA-binding proteins by their ability to bind to specific oligonucleotide probes.
The usefulness of this technique is hampered by its requirement for relatively large amounts of nuclear proteins (typically 50-100 mg), problems with protein degradation during isolation, and the difficulties in achieving efficient electrophoretic separation and transfer of a wide molecular size range of proteins.
A number of modifications to the Southwestern blotting was done to increase the sensitivity of detection without excessive background and with the improved ease of room temperature hybridization.

A Southern blot is a method in molecular biology of enhancing the result of an agarose gel electrophoresis by marking specific DNA sequences.
This caused other blot methods to be named similarly as plays on Southern's name (for example, Western blot, Northern blot, Southwestern blot).
Prior to blotting, the gel may be treated with a weak acid, such as dilute HCl, which acts to depurinate the DNA fragments.
