Nikon MicroscopyU: Specimen Contrast in Optical Microscopy
Specimen shading introduced by the compensator results in one side of the image appearing dark while the other side is brighter, leading to the perception of a pseudo three-dimensional image.
When considering transparent phase specimens that are poorly imaged in brightfield illumination, the role of the specimen in altering the optical path length (in effect, the relative phase shift) of waves passing through is of paramount importance.
A majority of phase specimens that are observed in culture vessels or sandwiched between a microscope slide and coverslip are relatively flat or plate-like as illustrated in Figure 4 for a hypothetical specimen immersed in a homogeneous medium.

Specimens must be received in the proper container to be accepted by the Laboratory.
Events which may lead to the rejection of a specimen include specimen improperly labeled or unlabeled, specimen improperly collected and/or preserved, specimen sample volume not sufficient for requirement of test protocol, outside of container contaminated by specimen or patient not properly prepared for test.
Specimens with requisitions that DO NOT contain required information (Dr.'s name, phone or pager #, diagnosis) will be held until the requisition is brought into regulatory compliance except in the case of life-threatening events.

Basics of Specimen Collection and Handling of Urine Testing
Random Specimen This is the specimen most commonly sent to the laboratory for analysis, primarily because it is the easiest to obtain and is readily available.
This specimen is usually submitted for urinalysis and microscopic analysis, although it is not the specimen of choice for either of these tests.
When a specimen is directly transferred from a collection cup into a preservative tube, it provides a stable environment for the specimen until testing can be conducted and reduces the risk of bacterial overgrowth or specimen decomposition.

AIDS Cancer Specimen Resource - Donate Specimens
The AIDS and Cancer Specimen Resource (ACSR) is a resource of HIV and non-HIV infected tissues and other specimens for use by qualified investigators to facilitate their research.
Blood specimens (one yellow and one red top tube) should be processed according to the ACSR guidelines, stored and shipped in batches directly to the ACSR using the same packaging and shipping guidelines as tissue.
A second reason for the double package is that the specimens should be shipped at room temperature; the packaging of the small shipping container is sometimes not enough to protect the specimens from hot or cold extremes.

Caused by an incident electron colliding with an atom in the specimen which is nearly normal to the incident's path.

This forces you to treat the specimen very gently in the course of removing water from it and coating it with the palladium/gold film which permits good SEM signals/images.
If the specimen is large (e.g., plant tissue, insect), epoxy cement is the best way; if the specimen is on a flat support (see later) one of these sticky tabs is a good solution.

 Type Specimen Catalog of Fungi
Work is underway to catalog the type specimens of fungi at the New York Botanical Garden Herbarium, as a continuation of the Type Specimen Catalog Project.
The type specimens of rust fungi (Pucciniaceae) were completed several years ago, and types from recently incorporated herbaria (e.g., Tulane University, Kansas State University) have also been cataloged.
Therefore, in addition to a transcription of the specimen label and an image of the specimen, type catalog records for fungi include text, images of associated unpublished notes and drawings, published descriptions and images of spore print, living organism, or microscopic features.
