Pauline Sperry was born in Peabody, Massachusetts to Willard Sperry, a Congregational minister, and Henrietta Leoroyd Sperry.
Sperry returned to Smith College for the 1916-1917 academic year as an assistant professor of mathematics, but moved to the University of California at Berkeley in 1917 as an instructor.
Sperry regularly taught the graduate courses in Differential Geometry, Metric Differential Geometry (the application of the calculus to the metric theory of twisted curves and surfaces) and Projective Differential Geometry.

In 1951, Sperry was invited to present his work at the California Institute of Technology (Caltech), which was seeking to fill the newly endowed Hixon Professorship of Psychobiology.
Sperry showed that if nerve connections were rearranged--for example, by redirecting to the other side of the animal the sensory nerves that innervate the left foot of a rat--inappropriate responses resulted that could not be unlearned.
Essentially, Sperry and his students showed that if the two hemispheres of the brain are separated by severing the corpus callosum (the large band of fibers that connects them), the transfer of information between the hemispheres ceases, and the coexistence in the same individual of two functionally different brains can be demonstrated.

In the early 1960s, Sperry and colleagues, including Michael Gazzaniga, conducted extensive experiments on an epileptic patient who had had his corpus collosum, the "bridge" between the left and right hemispheres of the brain, split so that the connection was severed.
At first the patient seemed quite normal, but experimentation showed certain activities such as naming objects or putting blocks together in a prescribed way could only be done when using one side of the brain or the other.
Sperry's work helped chart a map of the brain and opened whole fields of psychological and philosophical questions.

By the time Sperry had established himself as an accomplished inventor, a man named G. Hopkins had invented the first electrical gyroscope -- a disk mounted so that it remained in a fixed position despite the movements of its base.
