Spirogyra owes its name to a chloroplast (the green part of the cell) that is wound into a spiral, a unique property of this genus which makes it easily to recognise.
If we look at a filament of Spirogyra with the microscope, the first thing that attracts attention is the chloroplast, a narrow, banded spiral with serrated edges.
Spirogyra can also exhibit, apart from the ladder like conjugation, another form of conjugation.

Spirogyra is a filamentous green alga which is common in freshwater habitats.
In common with other members of its phylum (Gamophyta) Spirogyra lacks a motile variant at all stages of its life history; ie, no motile gametes (ova or sperm), no zoospores etc. Sexual reproduction is by a process called conjugation -- another of the famously remarkable sights available to the microscopist.
It looks like the stage in conjugation of Spirogyra in which the contraction of the cell contents to a ball is not quite complete, and the spiral nature of the chloroplast is still discernable.

Owing its generic name, Spirogyra, to the unique manner in which members of this genus twist their green chloroplasts into spirals, this key characteristic makes it easy to recognize species in this cosmopolitan genus of the phylum Gamophyta (conjugating green algae) in the division Chlorophyta.
Often an indicator of "nutrient enrichment" (actually, over-fertilization because of excess nitrate and phosphate pollution in contaminated stormwater runoff), freshwater ponds, ditches, vernal pools, and springs are often choked with tangles of this multicellular alga.
The zygospore is adapted to surviving harsh conditions, with a thick cell wall, and emerges as a new algal filament once the environment is more favorable, perhaps after overwintering in the sediments.

Sometimes alternatively known as water-silk, mermaid's tresses, or pond scum, a large presence of the unbranched algae often indicates the nutrient enrichment of freshwater bodies.
Spirogyra usually reproduce via vegetative means, individual filaments fragmenting to form new algal strands.
Adjacent filaments are capable of extending conjugation tubes toward each other in order to exchange genetic material and produce a zygospore, which is protected by a thick cellular wall that enables it to survive in sediment until more favorable conditions emerge.

The proposed work is designed as a contribution on the autecology and species delineation of Spirogyra including the compilation of a database containing morphological characteristics and ITS sequences.

 DNA Isolation and RAPD Analysis of Cladophora, Spirogyra, Bryopsis, with focus on Derbesia
However, attempts at attaining a genomic fingerprint utilizing the RAPD technique for the green alga Derbesia in comparison to those of Cladophora, Spirogyra, and Bryopsis were unsuccessful, possibly due to non-optimal RAPD-PCR conditions and erroneous reactant concentrations.
Even with a high purity sample of algae DNA (Bryopsis, ratio 1.75), no bands were detected in any of the trials ran, which is contrary to the expectations.
It is not surprising that the lesser pure DNA samples, Derbesia (1.65), Cladophora (1.68) and Spirogyra (1.69), generated no positive results either.

Spirogyra is a filamentous algae that can be found in almost every pond or ditch.
At the end of the summer Spirogyra and it's relatives Mougeotia and Zygnema grow to such numbers that they form a thick scum on the surface of the water.
Like their relatives the desmids, they belong to the phyla Gamophyta, the conjugating green algae.
