The deepest layer of the epidermis is the stratum basale.
The stratum spinosum (SS) and stratum basale (SB) are labeled (note the melanin in the latter!) and the stratum granulosum (not labeled) is visible in the left-hand image.
This is the stratum corneum or "horny layer." It's the actual interface between the outer surface of the animal and its environment, the layer from whence cells are cast off in the normal process of wear and tear.

Nearest the outer surface is a layer of cornified dead cells (stratum corneum); next is the granular cell layer (stratum granulosum); next the pentagonal cell layer (the stratum spinosum); and finally the undifferentiated cells stratum germinativum.
The lower density of gap junctions in the stratum granulosum raised the possibility that the cells in that layer did not exchange ions with the neighboring strata.
The zero-time gap junction density of the stratum granulosum found in this study (0.041 ± 0.013) is not significantly different from the gap junction density (0.04 ± 0.018) found previously (6).

The epidermis may further be divided into the following four or five layers (from the top down): the stratum corneum, the stratum lucidum (in the lips and thickened skin of the soles and palms), the stratum granulosum, the stratum spinosum, and the stratum basale.
Stratum spinosum is made of several layers of cells that stain with spine-like extensions; these cells have limited mitotic activity.
Stratum basale is a single layer of cuboidal cells in contact with the basement membrane.

The cells of the stratum granulosum that surround the developing ovum form a layer called the corona radiata.
Outside the stratum granulosum is a two-layered structure called the theca.
The theca interna (next to the stratum granulosum) contains large, glandular cells and is responsible for secreting androgens that will be converted into estrogens by the granulosa cells.

Keratinisation should not be used as a synonym for the formation of the stratum corneum: other stratified squamous epithelia may become keratinised but may not form a stratum corneum in which cells join to form a horny cell membrane.
The stratum granulosum is formed by a single layer of very dark and flattened cells in thin skin.
The stratum basale is formed by a single layer of cuboidal or columnar cells and delimits the epidermis from the dermis.

The stratum spinosum (spino = thorn, prickle/lat.), the prickle-cell layer, is above the basal layer.
Above the prickle-cell layer is the stratum granulosum (granula = grain/Lat.), where the cornification (keratinization) of the keratinocytes begins.
The stratum corneum (cornea = horny skin/Lat.) is the uppermost layer of the epidermis.

This is supported by the fact that the stratum spinosum is thicker in portions of skin subject to frequent abrasion such as palms and soles.
Since the upper layer of skin is heavily keratinized and the lower layer (stratum germinativum) is not, it is clear that as cells of the epidermis progress toward the external surface, one of the things that is happening is that higher and higher concentrations of keratin are being synthesized and deposited in the cytoplasm.
The hair follicle, in a sense, is a deep invagination of the epidermis that involves the stratum basale and stratum spinosum.

As the keratinocytes move upward and differentiate, keratin filaments (a type of intermediate filament) aggregate to form larger groups of filaments (tonofilaments) and even larger tonofibrils which are visible by light microscopy in the spinosum layer.
During differentiation, in the stratum granulosum, aggregations of keratin filaments become embedded in matrix proteins derived originally from keratohyaline granules.
In the cells of the stratum corneum, this keratin protein complex occupies the entire dead cell.

The outer most layer of cells is known as the stratum corneum, the stratum lucidum directly under the stratum corneum, then the stratum granulosum, the stratum spinosum and the deepest layer of cells in the epithelium is the stratum germinativum, sometimes referred to as the basal layer.
The replacement process is normally an orderly movement of cells from the stratum granulosum upward to the stratum corneum.
On the way through progression of the four upper layers, the cells leave their nutrient supply behind, produce keratin, lose their nuclei and granules and die, forming scales in the outermost hard surface on the skin.
