		The morphology of the T-tubules was examined using confocal microscopy and an extracellular fluorescent dye, sulforhodamine B. In addition the accessibility of the T-tubules from the extracellular space was evaluated from the rate of washout of sulforhodamine B from the T-tubules.
		Sulforhodamine B fluorescence was excited using the 543 nm line of a helium-neon laser operated at 50 % maximum power and emitted light was collected between 570 and 610 nm.
