Buffett, chairman of Berkshire Hathaway and a hugely influential investor, was with his wife when she died in Cody, Wyo., where they were visiting friends, the company said.
Susan Buffett, a Berkshire director, was listed this year by Forbes magazine as the 60th-richest American, with a personal net worth of $3.1 billion.
The foundation's secretary, Allen Greenberg, said Susan Buffett's legacy will be her work as a champion of civil rights, reproductive health and AIDS awareness.

OMAHA - A private funeral restricted to family will be held for Susan Buffett, the wife of billionaire investor Warren Buffett, one of her sons said Friday.
Susan Buffett died Thursday of a stroke at age 72 while she and her husband were visiting investor Herbert Allen of New York at his ranch in Cody, Wyo.
Peter Buffett said he flew to Cody and was at his mother's side when she died at the local hospital, as were his father and his sister, Susie.

Despite his immense personal wealth, Buffett has not been particularly charitable -- even now, late in his life, a time when many moguls, with their eyes on the history books, seem suddenly to develop an urge to share.
But the Buffett Foundation, which was established in the mid-1960s, disburses a pittance of his wealth -- just $11 million to $12 million a year, mostly to family-planning clinics.
Although he has three children, his wife is his sole heir, and Buffett has said that he intends for 99 percent of his money to eventually go to his foundation, which will likely become the largest endowment in the country.
