 Transposon - Wikipedia, the free encyclopedia
Transposons are sequences of DNA that can move around to different positions within the genome of a single cell, a process called transposition.
Both classes of transposon may lose their ability to synthesise reverse transcriptase or transposase through mutation, yet continue to jump through the genome because other transposons are still producing the necessary enzyme.
While transposons may confer some benefits on their hosts, they are generally considered to be selfish DNA parasites that live within the genome of cellular organisms.

After the transposon is ligated to the host DNA, the gaps are filled in by Watson-Crick base pairing.
Often transposons lose their gene for transposase; but as long as somewhere in the cell there is a transposon that can synthesize the enzyme, their inverted repeats are recognized and they, too, can be moved to a new location.

The term"selectable marker"refers to a gene that, when carried by a transposon, alters the ability of a cell harboring the transposon to grow or survive in a given growth enviroment relative to a similar cell lacking the selectable marker.
For example, the selectable marker gene within the transposon of the invention may influence the protein expression when a construct obtained by the method of the invention is inserted into a protein expression plasmid.

A population of cells that each contains transposon insertions at random positions in the genome (a transposon "pool") can be used to select or screen for those cells with insertions in or near a particular gene.
To isolate insertions in or near a gene, cells with transposon insertions are first selected by plating onto rich medium containing an antibiotic resistance expressed by the transposon.
