Triclabendazole is given, after consumption of food, in a single dose of 10 mg/kg body weight.
Different doses of triclabendazole were studied during these trials and it was concluded that the most efficient dosage for fascioliasis was 10 mg/kg body weight, while for paragonimiasis it was 20 mg/kg, given in two equal doses.
Following the registration of triclabendazole for human use and noting its use for public health purposes, the WHO Expert Committee on the Use of Essential Drugs recommended, at its December 1997 meeting, that the drug be put on the list of essential drugs.

The pharmacokinetics and flukicidal efficacy of triclabendazole delivered in low doses on a daily basis through urea molasses blocks were studied in cattle and buffaloes experimentally infected with Fasciola gigantica.
After receipt of triclabendazole by cattle and buffaloes at 0.83 and 3.0 mg/kg body weight, respectively, on a daily basis, both the animal species exhibited equilibrium between its absorption and disposition of its metabolites in plasma on day 4 and remained almost unchanged thereafter.
The continuous subtherapeutic dosing of triclabendazole through urea molasses block at 0.83 mg/kg body weight in cattle and 3.0 mg/kg body weight in buffalo proved to be efficacious against mature liver flukes.

Triclabendazole in the treatment of Paragonimiasis skrjabini
Triclabendazole as a chemically pure powder was given orally in the dosage of 10 mg/kg bid for 3 days.
In the second experiment, the dose of triclabendazole was increased to 150 mg・kg(-1)・d(-1) and 200 mg・kg(-1)・d(-1), for 3 days with a total dosage of 450 mg/kg and 600 mg/kg, respectively, and worm reduction rates of 80.8% and 86.7%, respectively.
The mechanism of action of triclabendazole may be due to inhibition of protein synthesis of flukes[10] leading to protein depletion, generalized atrophy of internal organs and death.Clinically triclabendazole is highly effective in treatment of various forms of P. skrjabini including pulmonary, pleural, cerebral and subcutaneous tissue lesions.

The influence of triclabendazole treatment on the pathophysiology of the disease, in terms of haematological parameters and serum enzyme levels, and in the dynamics of production of specific antibodies to excretory/secretory products (ESP) of F. hepatica were also examined.
In those animals in which triclabendazole was highly effective, antibody levels fell back to negative values similar to those recorded preinoculation at 18-21 weeks PI.
