Topic: Tropoelastin

  Patent 6,808,707
The composition comprises virgin monomers of tropoelastin and lysyl oxidase.
The notion that tropoelastin monomers are secreted from the cell and diffuse onto the surface of growing fibers seems to be inadequate to explain the efficiency of the whole process and the variable forms of elastic fibers in different tissues.
Since tropoelastin monomers are normally secreted from the cell, a preferred way to produce the monomers for the present invention would be to introduce the gene with the signal sequence intact and under the control of a strong promoter into a cell and grow the recombinant cells in culture.

Hydrophobic Domains of Human Tropoelastin Interact in a Context-dependent Manner -- Toonkool et al. 276 (48): 44575 -- ...   (Site not responding. Last check: 2007-11-02)
Tropoelastin is the soluble precursor of elastin, the major component of the extracellular elastic fiber.
The plots in a and b are representative of the coacervation profiles of all five tropoelastin constructs, albeit the individual construct profiles varied because of differences in coacervation behavior as shown in panel c and Table II.
Domain 26 of tropoelastin consists of four nonapeptide repeats of the sequence XGXGXGVPG, where X is either A, L, V, or I. It plays a dominant role in coacervation (15).

Elastic fibers, consisting of soluble tropoelastin monomers cross-linked on a preexisting scaffold of microfibrils, are produced primarily during late fetal and neonatal stages of development.
The onset and inductive phase of tropoelastin expression are characterized by increased transcription of the tropoelastin gene.
We have found in silica-induced pulmonary fibrosis that expression of tropoelastin is primarily increased in the walls and the septal tips of the alveolus, with modest increases in other compartments which normally express tropoelastin during development.

News in Science - Can damaged tissues spring back into shape? - 13/09/2002
Called tropoelastin, these building blocks, like bricks in a brick wall, are cemented together to form elastin.
The team first discovered how to make tropoelastin in the mid 1990s and last year was awarded a US patent on the synthesis and cementing process.
The shapes are made by placing the tropoelastin 'bricks' and the 'cement' in moulds to determine the shape, and 'cooking' the mixture at body temperature for 10 minutes.

The present work explores some of the properties of the central hydrophobic domains of tropoelastin and their contribution to association in the context of position in the full length molecule.
Various tropoelastin constructs, including a naturally occurring form of tropoelastin, its mutant derivatives, and shortened molecules, were studied with respect to their secondary structures and association abilities.
These results indicate that the assembly of tropoelastin molecules is not merely a result of random interactions of hydrophobic domains but that it is caused by multiple and specific interactions of individual hydrophobic domains.
