Identification and genomic characterization of a new virus (Tymoviridae family) associated with citrus sudden death disease.
The family Tymoviridae comprises the genus Tymovirus, from which it derives its name, the genus Marafivirus and the newly established genus Maculavirus.
Two histidines of the coat protein of turnip yellow mosaic virus at the capsid interior are crucial for viability.

Tymoviridae are monopartite single-stranded positive sense RNA plant viruses, belonging to group IV of the Baltimore classification of viruses.
The genome consists of one large open reading frame and is capped at the 5’ terminus.
The Tymoviridae are a relatively new family, only approved in 2002.

This family of viruses is comprised of three genera known as the genus Tymovirus, the genus Marafivirus, and the genus Maculavirus.
Such classification and description of viruses is useful for comparison with other known and unknown viruses and for prediction of virus properties.
Technical Abstract: The family Tymoviridae comprises the genus Tymovirus, from which it derives its name, the genus Marafivirus and the newly established genus Maculavirus.

Subsequences representing the conserved region according to the PSI-Blast alignment, corresponding to residues 35–378 of the query sequence, were aligned using ClustalX, and a Pfam type profile was generated and calibrated using tools from the HMMER package.
The intervening regions between the conserved domains are extremely variable in these sequences, and this tended to confuse alignment programs in the sense that conserved regions were not correctly aligned.
A Pfam-type profile for the methyltransferase domains of Flexiviridae and Tymoviridae was generated from a ClustalX alignment, using hmmbuild and hmmcalibrate from the HMMER package.
