Yarra Glen had a hotel, post office and primary school by 1870 and numerous surrounding holdings which ran stock or had vineyards for wine production (the Ryrie brothers had planted vines in 1838).
Yarra Glen is situated a little north of a flood plain through which the Yarra River flows.
However, when bridges over the Yarra River were built the route along Whitehorse Road to Lilydale was more convenient than the Yarra Track over Christmas Hills and through flood-prone Yarra Glen, and traffic tended to avoid the area.

The Terminus was the first railway station in Australia, and the opening day saw the first mechanical train trip in the country's history.
Melbourne's 16 radial electrified train lines all originate from the station.
The first railway building to occupy the Flinders Street site was simply called Melbourne or City Terminus, and was a collection of weatherboard train sheds.

Yarra Glen is located in the heart of the fertile Yarra Valley which is noted, above all things, for its wineries.
One of Yarra Glen's more impressive buildings is the Grand Hotel, at 19 Bell St. Built in 1888, during the boom period, this two-storey stuccoed hotel has a four-storey tower at one end, a masonry arcade along the street frontage, a cast-iron verandah and a parapet.
The Yarra Glen Craft Market is held at the same venue on the first Sunday of the month from October to June (in March it is on the third Sunday), tel: (03) 5988 4424.

 Healesville Line
The Yarra Glen station building remains in situ, however this building was partially destroyed by fire several years ago and much restoration work is needed.
Operations from the Yarra Glen end of the line have not been as regular to date, although the level crossing at Melba Highway has been restored with flashing lights.
Due to the high costs associated with the section of track between Coldstream and Yarra Glen because of the condition of the bridges, they discontinued their lease of this section and instead concentrated their efforts in the section between Healesville and Yarra Glen.
