 Cell Isolation Theory
On the lateral surfaces of adjacent epithelial cells there are four distinct types of intercellular bonds: the zonula occludens, zonula adherens, macula adherens and nexus.
In the zonula occludens, or "tight junction", there are multiple sites of actual fusion of the adjacent unit membranes interspersed by short regions of unit membrane separation of approximately 100-150 Å.
In a zonula adherens, or "intermediate junction", a fine network of cytoplasmic filaments radiates from the cell membrane into the cytoplasm.

  Tjp1 - Tight junction protein ZO-1
Colocalization of Cx43 and of the tight-junction- associated protein zonula occludens 1 (ZO-1) was demonstrated in seminiferous tubules by using dual-label immunofluorescence in conjunction with confocal microscopy.
Zonula occludens proteins (ZOPs), currently comprising ZO-1, ZO-2, and ZO-3, belong to the family of membrane-associated guanylate kinase homologue (MAGUK) proteins that are involved in the organization of epithelial and endothelial intercellular junctions.
To this purpose, we used two tight junction markers, the Zonula occludens (ZO-1) and claudin-1 proteins, and a glial marker, the aquaporin-4 (AQP4) protein, whose expression is correlated with BBB differentiation and integrity.

One of the tight junction components, zonula occludens protein 2 (ZO-2), is expressed as two isoforms, ZO-2A and ZO-2C, in normal epithelia.
We have observed that 2 forms of zonula occludens 2 (ZO-2) protein, ZO-2A and ZO-2C, are expressed in normal human pancreatic duct cells, but only ZO-2C in pancreatic duct adenocarcinoma.

These particular vessels displayed an increase in phosphotyrosine staining, loss of the tight junctional proteins, occludin and zonula occludens-1, and apparent redistribution of the adherens junction protein vinculin.
Our results have established an endothelial cell molecular profile associated with leucocyte-induced blood-brain barrier breakdown in vivo, and the relevance of different in vitro cell culture models may now be viewed more objectively.

 III. Tubular Anatomy and Function
Around each cell near its luminal or apical face is a circumferential belt, the zonula occludens (tight junction) which connects it with neighboring cells.
The width of the zonula occludens and the paracellular space have been greatly exaggerated for purposes of emphasis.
The zonula occludens surrounding these cells has a low permeability to Na so there is little passive diffusion back into the lumen and the cells can reduce the urine Na concentration to very low levels.

 Epithelium
zonula adherens, which also completely encircles each cell, but the adjacent plasma membranes are separated by a gap of about 20 nm.
While it is the zonula occludens of the junctional complex that principally affects this function, it is the adhesive properties of the zonula and macula adherens which guard against physical disruption of the barrier.
Cell-cell and cell-extracellular matrix (ECM) adhesive interactions are clearly important in maintaining the integrity of epithelial sheets, and in some disease states--most noticeably inflammation, tumor invasion and metastasis (the spread of cancer cells from one site to another).

Researchers at the Center for Vaccine Development at the University of Maryland were trying to develop a vaccine for the cholera bacterium (cholera is one of the leading causes of death in children worldwide, and it causes a profuse, watery diarrhea).
The researchers discovered that cholera causes diarrhea by secreting a toxin called Zot (zonula occludens toxin), which can open the tight junctions (zonula occludens), thereby contributing to the severe, life-threatening diarrhea.

Zonula occludens form a complete, tight seal and prevent the passage of material across the epithelium (e.g., macromolecules are prevented from passing from the intestinal lumen into the intercellular space).
Together, the zonula occludens, zonula adherens, and macula adherens constitute the junctional complex visible in the electron microscope.
The microfilaments from the microvilli and zonula adherens, as well as the tonofilaments from the desmosomes, all intermingle to some degree within the terminal web.

Age-related changes in rat cerebral occludin and zonula occludens-1 (ZO-1).
Since aging is associated with significant alterations in the blood-brain barrier (BBB) it is possible that specific tight junction proteins may be altered in the cerebrum of aging rats.
To test this hypothesis, Western and Northern blot analysis were carried out to measure the steady-state level of occludin and zonula occludens-one (ZO-1) proteins and their mRNA in cerebral tissue of 3-, 12- and 24-month-old rats.

A systematic analysis of 40 random genes in cultured vascular smooth muscle subtypes reveals a heterogeneity of gene expression and identifies the tight junction gene zonula occludens 2 as a marker of epithelioid "pup" smooth muscle cells and a participant in carotid neointimal formation.
The following 5 consistently differentially expressed genes were identified in vitro: zonula occludens 2 (ZO-2); peroxisome proliferator-activated receptor delta (PPARdelta); secreted protein, acidic and rich in cysteine (SPARC); alpha1(I)collagen; and A2, an uncharacterized gene.
We examined these 5 clones during carotid artery injury and an inconsistently differentially expressed clone Krox-24 because, as an early response transcription factor, it could be involved in the injury response.

The terminal bar (arrow), visualized as dark spots that punctuate the terminal web at the point where two cells touch at their apical margins, marks the location of the apical junctional complex.
This complex consists of the zonula occludens junction, which forms a water tight seal around the apex of the columnar epithelial cells and the zonula adheres junction, which attaches adjacent cells firmly together.
The zonula adherens junction is reinforced by a band of actin microfilaments located on the cytoplasmic face of the plasma membrane.
