Earlier experiments had shown that urinary elimination of phenylethylamine (PEA), a metabolite of phenylalanine, is decreased in some cases of depression and therefore, Yaryura-Tobias believed that the administration of the phenylethylamine precursor, phenylalanine might improve cases of depression.
The authors hypothesize that a reduction in brain levels of phenylethylamine (the phenyalalnine metabolite) was one factor in causing the depression.
At the start of the study, the depressed patients had significantly lower blood and urine concentrations of phenylacetic acid (PAA), a metabolite of phenylethylamine (which is a metabolite of phenylalanine) as compared to the 48 control subjects.

Phenylethylamine has always been the darling of the pharmacologist in that it is structurally clean — it is naturally present in human fluids and tissues - and because it has a close chemical relationship to pharmaceutical stimulants.
Phenylethylamine is also found in chocolate, and is responsible for its effects on mood, appetite and sense of well being.
Phenylethylamine is intrinsically likely the cleanest stimulant known to man. Its ability to stimulate the central nervous system without causing jitters or nervousness is extraordinary!

Phenylethylamine is known as the "love drug" and is thought be be the reason why chocolate is said to be an aphrodisiac.
It is a chemical that mimics the brain chemistry of a person in love, so when levels of phenylethylamine are high in the body it relieves depression from unrequited love.

Phenylethylamine is an endogenous neuro-amine that has been linked to the regulation of physical energy, mood, and attention.
There is evidence that phenylethylamine and phenylacetic acid levels are very low in the biological fluids of depressed patients.
Administration of phenylethylamine or its precursor L-phenylalanine, in conjunction with selegiline, a selective monoamine oxidase B inhibitor, has been reported to alleviate depression and to produce improvements in mood.

The team tested urine samples on their "day of rest", analyzing for phenylacetic acid, a by-product of turnover of the endogenous neurotransmitter phenylethylamine, involved in the brain's activity during exercise.
Previous research has simply measured levels of phenylethylamine itself but the researchers believe their approach provides a more accurate assessment of the compound's activity in the body.
However, they point to the structural similarity of phenylethylamine to the amphetamine drugs as a possible cue to the "high" felt by runners, a phenomenon linked to natural endorphin activity in the brain.

For 18 of the 20 men, urine phenylacetic acid--which reflects phenylethylamine levels--was higher after exercise, increasing anywhere from 14% to 572%.
Billett and colleagues suggest that the effects of exercise on phenylethylamine levels be tested in people with clinical depression.

Phenylethylamine, however, is not stored in intra-neuronal vesicles and is rapidly degraded by monoamine oxidase-B. Despite its short half-life, phenylethylamine attracts attention as an endogenous amphetamine since it can potentiate catecholaminergic neurotransmission and induce striatal hyperreactivity.
Excess phenylethylamine has been invoked particularly in paranoid schizophrenia, in which it is thought to act as an endogenous amphetamine and, therefore, would be antagonized by neuroleptics.
The importance of phenylethylamine in mental disorders is far from fully elucidated but the evolution of phenylethylamine concentrations in relation to symptoms remains a worthwhile investigation for individual psychotic patients.

Vitamin B6 activates the enzyme that converts phenylalanine to phenylethylamine, so the combination of low-dose deprenyl, B6, and phenylalanine is another biological way to enhance both phenylethylamine and catecholamine brain function, and thus diminish depression.

Now it appears that the antidepressant effects may be due to a chemical called phenylethylamine.
The study required that they stop exercise completely for a day, during which their urine samples were measured for phenylacetic acid, a by-product of phenylethylamine turnover, and a more accurate measure of the chemical.
There may be many factors involved in the phenylacetic acid response to exercise, say the authors, but considering that its chemical structure is very similar to that of amphetamines, it may be that this chemical is part of a “runner’s high,” a phenomenon linked to natural endorphin activity in the brain.
